The results of analysis for infectious agent markers in cord blood samples intended for the public register using IFA and PCR

In accordance with the Ministry of Health's Order no. 325 “About the developing of cells technologies”, 25 June 2003, each cord blood sample should be tested for the presence of infectious agents by testing for the infectious agents’ antigens or antibodies: anti-HIV 1 and 2, p24 HIV1, anti-HTLV I and II, anti-НВcorAg, HBsAg, anti-HCV, anti- CMV, anti-Toxoplasma gondii and RW, and for the presence of microbiological contamination by Staphylococci, Streptococci, Enterococci, Neisseria gonorrheae, Candida species, Aspergillus species, and Esherihia coli.

The ELISA and ID-PaGIA methods were used for the identification of the infectious agents’ markers in the plasma of cord blood samples. The systems adopted for aerobic and anaerobic bacterial and fungal growth were used for microbiological contamination analysis. In the case of the identification of aerobic bacteria, an antibiogram was used.

In 2.2% of samples antibodies to HBcorAG were detected, in 0% antibodies to HCV, in 67% antibodies to CMV, in 8.3% antibodies to T.gondii, and in 16.9% both antibodies to CMV and T.gondii were detected. In 3 cases antibodies to T.pallidum were detected using ID-PaGIA, but the RW was negative. It was noted in the patients’ medical history that T.pallidum had caused their disease. Antibodies to HTLV I and II were not detected. No infectious markers were detected in 26.8% of tested cord blood samples.

No pathogenic microflora and no fungi were detected in the samples. Semi-pathogenic microflora was identified in 7.2% of samples. An antibiogram was made for all detected aerobic bacteria so that the cord blood sample was suitable for transplantation.

According to the above-mentioned data, 1400 cord blood samples for public use were selected for storage in the “Stem cell bank Pokrovsky”. These samples do not contain antibodies to HIV-1 and -2, HIV-1Ag, HTLV-I and -II, HbcorAg, HBs-Ag, or HCV and correspond to the Ministry of Health's Order no. 325.
